This 'R' sign indicates the road is part of a ring road. Great Eastern Street is part of London's inner ring - see Image The road is now two way, as is Old Street (eastern part) and Shoreditch High Street. Previously all three were all one-way roads forming a one-way system where the traffic moved clockwise around the 'Shoreditch triangle'. [I am grateful to Peter Thwaite for this information.]

Former electricity generating substation, Rivington Street, EC2
The plaque on the wall (just to the left of the white doorway) says that the building was a transformer station for the tramway system; it was designed by Vincent Harris, an architect with London County Council, and built in 1905-7.
The building is currently occupied by the Tramshed restaurant and the CͶB Gallery.
